Objective To determine the contents of luteolin and hesperidin in the effective fractions of Spica Schizonepeta by using HPLC.Methods The contents of luteolin and hesperidin was determined at indoor temperature. The chromatographic conditions included TECHSPHERE ODS column (250mm×4.6mm
5μm) and the mobile phase consisted of acetonitrile-0.2% phosphoric acid running in a gradient mode with a flow rate of 1.0ml/min. The detection wavelength was at 283nm. Results The linear rangers for luteolin and hesperidin were 0.036~0.12ug (r=0.9999) and 0.27~0.9μg (r=0.9999) respectively. The average recovery of the procedure was 100.93%
RSD=1.59%(n=6)for luteolin
and 98.15%
RSD=0.43% (n=6) for hesperidin. Conclution This procedure
which is simple
accurate and feasible
can be used as a qualitative control of the effective fractions of Spica Schizonepeta.
